Heyo,

You can do some teams with all three of them, here are some ideas :

With Roshi :

Roshi - Fuguki Edo - Water Main - Sakura Cloak
2
This is a very classic team that is being used by many players. You can trigger roshi almost every round as long as your opponents are hitting Fuguki. Also, they are both scaling really decent overall, they get more tanky and Roshi get's a ninjutsu increase. The healing support plus the potion are keeping them long enough on the field so they can cause some trouble. You can additionally change Roshis position with Fuguki's if you happen to have a faster position 1 than your opponent.

Roshi - Yagura - Earth Main - Sakura Christmas
3
This team is also solid if you wish to try something else out. You c*so trigger Roshi here plus with Yagura you can copy any other opponents lineup ninja. This team works awesome as wind main as well, with the flower guard buff, which will make your Sakura immune. Another option here is to also run Sakura cloak, Gakido, Ningendo, Kurenai or any other supports that can be solid here. Fire main and water main do work as well here.

With Ay :

Ay is basically a ninja that needs chakra and only works with specifically teams, which are the following :

4
This is one of the classic teams with Ay, he needs Samui in the team, it's a must for the chakra. Ay is always a bursting character and works solid for the early stages, he is easy to counter however. This team is also working as wind main team or you can change Omoi with Darui as well. Lightning main is a third option as well here. So basically, Ay is working only with those formations, but his damage can be really great.

With Han :

Han has many combinations, he is a crucial ninja many times especially with that buff removing he can cause a lot of trouble. So you can make easier teams, you should think about what happens when a ninja removes the buffs of the opponent. Two things : 1) You cannot be bursted by same power levels or a small cap and 2) You can throw to them as many debuffs as you want, of course that works if they don't have too many heals and cleanses. Anyway, one option is the above shown with water main which is really nice, heals, survival, potion stacks, ignition. Other ideas can be the following :

Han - Mei - Sakura Christmas - Fire Main
5
This team can really be a trouble sometimes, you got a reflection, your Han will remove all the buffs, Mei bring up her barrier and the buff stays. Sakura is an additional safety here. This team works great with Hanzo as well, instead of Sakura. The Bani chakra passive can be changed always with the clone or with the Death Mirage, if you see opponent players running mostly with ninjutsu damaging ninjas, this passive can be really handy. Your Sakura summoning trees and healing is a decent support as well.

Han - Mei - Sakura Christmas - Water Main
6
The same team as above almost, but as position one water main. Basically here, you remove all the buffs and your water main is ready to go for a chaos fest. Nice combos coming out from this team, immobile from Mei in the chases. I would suggest you to try here Kurenai as well, instead of Mei or Sakura, this team can works with infinity combinations by having the water main and Han as main ninjas in the team. Such are : Gakido, Ningendo, Kurenai, Sakura GNW, Jigokudo, Naruto Suit.

I hope It helps!

NO. 5Tailed Han is a support type character, Edo Han is a blitz type character. They are entirely different and CANNOT replace one another.

Exactly as MonkeyLord said, nope, you really cannot here. It's really crucial when he can remove buffs. I can give you some Edo Han teams as well though if you need.
