After seeing some of the guides on this forum and on Youtube, I came to the conclusion that the Water Main guides take to long to achieve the desired outcome, so here is my take on it, taking bits and pieces from other guides dedicated to other mains.

IMPORTANT: requires level 65 for the chase talent but so does unlocking the difficult mode, may make it unavailable for the normal and easy mode or you can try "Shank Bomb Jutsu" as a chase and Kyushoku Deer summon.

ROUND 1 (Poison Giants - 3 turns):
- with Neji acupuncture the bottom giant, after the giants attack summon Sai's dogs
- after the giants attack heal up with MC mystery, wait for Sasuke to attack with standard and queue barrier to prepare for next round
- there should be only one giant left so let him attack and bring 2 more dogs from Sai (in case there are still 2 the top one sould die with 1 standard)
- done here

ROUND 2 (Shizune - 2 turns):
- immidiately acupuncture her with Neji and wait, Sai will start combo and you should have 4 dogs alive by this point but you can do with less (at least 2)
- before she can heal use Sasuke's Kirin mystery on her to finish her off and queue the barrier for next round
- done here

ROUND 3 (Karin - 2 turns):
- take the hit from the poison and her standard attack, heal up with MC mystery and bring 2 more dogs from Sai
- before she can do anything you need to acupuncture with Neji and immidiately use Sasuke's Kirin on her and queue the barrier for next round
- done here

ROUND 4 (Jiraiya - 2/3 turns): you can auto this part if you want to
- let Jiraiya kill one of the dogs and then bring up 2 more
- acupuncture with Neji, use Sasuke's Kirin and let Jiraiya attack with standard before healing up
- depending on your power he may need another turn of standard attacks if not you are done here

ROUND 5 (Hanzu - 2/3 turns):
- if you used a 3rd turn on Jiraiya you may use Neji mystery here for extra damage, don't bother with Sasuke barrier bring dogs only after Hanzu attacks
- after hanzu attacks heal up
- if he isn't dead already, immidiately acupuncture with Neji before he can cast the poison, he should die by the end of the turn or start of the next

And there you have it, can't post the points i got because i already had my six weekly runs done, and was just preparing for next week since I changed mains this week.
