With what you have you can make a variation of the Empress Build and use Kurenai, Shizune and Hinata with the passive that boosts female ninjas. If you get Karin switch Shizune for her.

Another possibility is Hinata, GNW kankuro and Sasuke(preferably 3* though but 2* still connects your combo)


-Best healer between Shizune and kabuto imo is Kabuto. Neither are impressive healers but his clones and Human experiment passive give him many uses like Strong Approaching or Ranked teams with Curse seal ninjas.
-Anko generally doesn't give any benefits to a wind main so unless you need her skills in your lineup then no, she would not be good. She's built for Fire mains.
-Tenten is good but again not that much for your main, the one that utilizes her best is Water with Poison Tai. GNW Tenten will be a different story once she's here.
-I don't really know much about Wind mains so can't tell you who to focus on getting or upgrading beside Sasuke and Naruto that "evolve" at 3 stars again, making them much more useful than they are now. But also keep in mind at level 60 you'll be able to change your main as well once a day so try planning ahead on what kind of team you want and start working towards it.
-Again, this is mostly dependant on the lineup you are gonna be using, and the mode you're in. Expect to change them around a lot, different setup for SA, different for 9tails, different for PvP etc
-They're quite different. Hinata's better as a tank and carries much more bulk but Neji's skill is also very useful at times. Especially in SA where interrupts are important he is used a lot if you don't have Tayuya. Or in Survival Trial where you know what's coming you c*e Neji to your advantage. the typical is:

Kurenai Main x
Karin/Shizune x Hinata
x x x

Or if you go with the other team kankuro where Kurenai goes and Sasuke where the Healer is. But make sure there's a clone in front of Main so Kankuro's puppet doesn't end at the front
