Since her passive is for the first mystery used, you c*e hashirama's mystery first to trigger the passive then use kushina. You could also add rin to counter your opponent.

Remember that rin's passive works for each unit in the team separately, meaning that if the unit didn't got hit by hashirama they will still have the 90% reduction on kushina's mystery. If you want max damage on kushina you will need another ninja that can do damage to all units, or just comform with high standard damage or sniping the most important units on rd 1.

So just use hashirama on Rin then kushina until you get rid of her? You don't want her to be alive so why target anything else?
