Hello.
The chakra points that you have while casting her mystery do count for her passive. When you have the default chakra on round 1 (20) and you cast her mystery which costs 20 chakra as well, you‘ll have 0 chakra left thus she won‘t have any scaling in that way.
PS: Correct, without using her mystery on round 1 you have 20% combo and ninjutsu buff.

When we click he mystery, the chakra is take before she hits anyway with her mystery. So when she cast her mystery, will her mystery be buffed by 20 percent and then the scaling is gone? or is it that there have to be 40 chakra and once she launches, the remaining 20 chakra afterwards is how much her mystery will be buffed once she hits the enemy?
