Hello.


The chakra points that you have while casting her mystery do count for her passive. When you have the default chakra on round 1 (20) and you cast her mystery which costs 20 chakra as well, you‘ll have 0 chakra left thus she won‘t have any scaling in that way.


PS: Correct, without using her mystery on round 1 you have 20% combo and ninjutsu buff.

Correct, when you have 40 chakra and launch her mystery, you'll have her 20% buffed basically in ninjutsu and combo, since her mystery costs 20 chakra to be launched. Let's say it more simple :

Round 1 : 20 chakra - Konan has there 20% buff in ninjutsu and combo, if you launch her mystery, her buff is 0% as you've got no chakra remaining.

Round 2 : 40 chakra - Konan has there 40% buff in the beginning of the round, when you launch her mystery there, you'll have 20 chakra points left, also a 20% buff in ninjutsu and combo.

Yeap or if in Great Ninja War or in some team mode if you're running for example a ninja that gives you chakra. If your teammate is faster and kills something while you clicked Konan you'll have 100 chakra there as well
