Changing any support ninjas does work with her on position one. Ningendo, Gakido, etc healing ninjas or supporting one can be considered as an option here.

What concerns Iruka, you can add him anywhere you wish, he does work. About Kushinmaru, he only does work with ninja that may buff edo tensei and the best option is lightning main also. He used to be a super fast nuker long time ago, but nowadays he's really easy to be countered. Most classic teams with him were teaming him up with Sasori Edo, Jinpachi (He is a must pretty much) and a barrier ninja.

What concerns Mei, you c*e her in any of the water main teams, for example :

If you wish to add instead of Gakido any female support or buffing ninja, you could do that. Also, Iruka for example would fit in that team as well.

Now, about Sasuke. To bring out his best potential you'll have to run him with mystery reset passives, like fire main's and lightning main's. Example lineups could be the following with him :

This is a full nuke sasuke team that works with lightning main as well, exactly the same formation. More ninjas to play around with him are for example Ino GNW, Shikamaru Chunin, Fu, or Darui as *ternative barrier ninja. Ninjas that can that c*l support/buff him do work with him. In case there are some buff removing ninjas, you can move main position 2 and run the seal on him to stop them for a while. There are more lineups with him, but Sasuke does need Edo Itachi to be a support next to him to actually work really solid.
