what is the version game we are currently in ? i check about say 2.4.0.1212 version
does the resistant / defense reduction a debuff ? how long it last ? does it stack up ?

yes the version is that one, but for the game content (events, ninjas, etc...) they call it 5.0 here.
about your second question, depends on which effect you are talking about.
Jiraiya, Ino, Kodumaru,Temari,sukura, karin they have decreasing enemies resistant .
how long does it last and can stack up ?

When not stated differently the effect lasts for 3 rounds (like the shields or poison) unless they are renewed.
About the fact if they stack or not depends on the specific effect.
