The reason I set things up this way is because Chiriku may get killed in round 1 if he takes on 4 ninjas worth of damage. The others have higher defence and resistance. Position 2 main is for debuff defence. Even if you remove the immunity with a mystery round 1 or 2, there's a status reflect to keep things in check. But if we're talking about any dodge ninja in round 2 that has higher.. even lower initiative, they might kill Chiriku early on.
the reason why he told you to place them that way is because in round 1 the top line hits are endured by konohamaru clone that will appear just over chiriku and if your enemy is fully in the bottom two lines then chiriku will be hit at most by 2 ninjas (never seen 3 ninjas formation in middle line), so is not a problem and chiriku will selfheal thanks to fire main passive.
