Too many things in this game is "heard" and there is no confirmation from Oasis. *ion runes are a great example, more players say it is so underwhelming and no one can explain what it does in terms of game mechanics. There should be some simple formula or % for what they do against defense and resistance but nobody from Oasis bothers to clarify.

He does not perfectly explains it, I came here from that video to do a research because there was no confirming answer. Just because he is a youtuber you shouldn't take everything what youtuber says as a fact unless really explained to why. He is basically saying the same thing what the guys are discussing here, that there is no actual proof at this moment whether or not these other attributes gives any kind of actual practical bonus or help.
BTrue thinks its true, he thinks people have done research, but he is basically basing his opinion on those people who have supposedly tested it, same people who are being mentioned here as well in above conversation if you would care to read. You see two sides conflicting about what whether it's true or not.
So BTrue is not explaining anything to why since he hasn't done a deep testing himself, he simply read about some stuff, maybe the same posts in which we are in and just read this one guys opinion or statement without much of a proof.
Poison damage and ignition damage depends only on enemy resistance vs attacking ninja ninjutsu, no other stat come in place for those two effects.
Damage reduction applies only to the damage dealt by standard attack/mysteries and works exactly like a flat increase of your defense/resistance.
If you have 9000 defense, 10000 resistance and 2500 damage reduction then you are considered like if you had 11500 defense and 12500 resistance for the damage dealt by standard attacks and mysteries.
Ninjutsu *ion and Taijutsu *ion come in place to reduce only the additional res/def you gained by damage reduction (so if you own an higher *ion that the enemy damage reduction stat all the excess of it is wasted).
For example, if your enemy ninja owns 1500 ninjutsu *ion and 2000 taijutsu *ion then, taking in account the previous example numbers i said, you are considered like if you had 9500 defense and 11000 resistance towards standard attacks and mystieries damage.

Here are some example tests:
this is the damage i normally endure against kakashi clone. The standard attack dealt 166-173, never went outside this range except very rare cases when kakashi did trigger his special effect for standard attack (causing knockdown) and then the damage was 121-128
this is the damage without my level 9 red damage reduction rune, this one:
the damage was always in the range of 192-199, except for the rare cases when kakashi procced the special effect (knockdown) and then was 142-149.
This stat works quite well.
interesting. I have also used the sand rune (because that is the only red one the game will give me) and have seen a very small damage reduction. however having something is better then nothing it's prob not the best choice over most of the other stat boosting runes that seem to be able to choose from.
