It says to use ninjas who can cancel debuffs, what are examples of it? I dont know unfortunately :( Also, what lineup would be best for this training for either wind main or water main? Read your ninjas, the ones that heal generally take off 1 layer of Debuff. Reading what your ninjas do honestly helps :x

Root teams can be made with light main if you have 3 sword ninjas in team your main can make them immune to buffs you c*e ninjas like sakura,tsunade,kabuto,and etc.

How do you cancel Guy's buff or the fire main's buff to his team? That's what makes this instance hard, not the cancellation of one particular character's mystery.

Wind is the most advantaged main for that lineup. Just use sand dust and be sure hidan is one of the targets. That way he will lose all the buffs and won't be able to land on you enough damage.
