Slot 1 > Slot 2 > Slot 3 > Slot 4. (If the skill is not "prompt" the standard attack will be replaced by mystery i.e.. prompt skill will always take priority over non-prompt skills.)

The way it stands your Main is move 1, Mifune move 2, Gaara move 3 & Mabui is move 4. If you're trying to outspeed someone that's on move 1 vs your Mifune @ move 2 it's never doing to happen unless you have the "sealed ninja" Ini lol.

The only way to find out before fighting someone if their Ini is higher is by asking. A simple trick to find out who has the higher ini while in a fight is to see who does their standard first and take notes.

Remember to also make use of the rebates when you're looking to refine your equipment.
