You can pretty much beat Hanzo on difficult with any main and the same team.

1 = Neji/Shurado/Any 0 battlefield cd, 2 cd ninja that can interrupt/acu/chaos/immobile on their prompt mystery
2 = Guy/Kabuto/main
3 = Guy/Kabuto/main
4 = Guy/Kabuto/main
Just make sure you have at least a 6-7 combo chase. For Lightning/Fire/Water, make sure you use a mystery that causes a chase. For Earth, make sure you take Earth Style - Super Lightened Boulder Jutsu. For Wind, take Dance of Impetus.
Vs Shizune:
For Water/Lightning/Fire, interrupt Shizune with your move 1 before she poisons. Make sure you have 3-4 Kabuto clones and combo after Shizune heals for her standard attack. Combo her with Guy/main's mystery. Next turn, tank Shizune's poison and use Guy/main's mystery to kill Shizune.
For Earth, use your mystery when you have 4 Kabuto clones. You should do 14 damage to Shizune with just standard attacks. Use Guy's mystery to kill her.
For Wind, do exactly like with Water/Fire/Lightning, but use Wind main's mystery on the same turn you used Guy's mystery. Queue on Guy's mystery for the next turn which basically puts Guy's mystery on a 2 CD.
Vs Karin:
Make sure you have 3-4 Kabuto clones and combo after Karin heals for her standard attack then combo her with Guy/main's mystery. Next turn, interrupt Karin with your move 1 because she heals before using her mystery. Use Guy/main's mystery to kill her.
For Earth, use your mystery when you have 4 Kabuto clones. You should do 14 damage to Karin with just standard attacks. Use Guy's mystery to kill her.
For Wind, do exactly like with Water/Fire/Lightning, but use Wind main's mystery on the same turn you used Guy's mystery. Queue on Guy's mystery for the next turn which basically puts Guy's mystery on a 2 CD. Also, make sure you interrupt Karin with your move 1 on the next turn with your move 1.
Vs Jiraya:
Jiraya uses his mystery on Turn 2, then every 2 turns after. Make sure you interrupt Jiraya with your move 1.
Vs Hanzo:
Hanzo uses his mystery on Turn 3, then every 2 turns after. Make sure you interrupt Hanzo with your move 1.
